John Cirrito, PhD
The Cirrito lab focuses on understanding the metabolism of amyloid-β (Aβ) within the brain extracellular fluid, or interstitial fluid (ISF). The lab developed a novel in vivo microdialysis technique that enables specific measurement of ISF Aβ in the brains of living and awake wildtype and APP transgenic mice.
Alzheimer’s disease (AD) is characterized by amyloid-β (Aβ) plaques and tau neurofibrillary tangles. Aβ accumulation initiates AD, with tau tangles contributing to neuronal death and disease progression. Aβ aggregates into toxic oligomers and plaques, leading to synaptic dysfunction and neuronal death. High concentrations of Aβ, particularly in the brain’s extracellular space (ISF), enhance aggregation. The lab utilizes microdialysis and micro-immunoelectrode (MIE) technologies to measure ISF Aβ levels in living mice.
